A protein with amino acid sequence homology to bovine insulin is present in the legume Vigna unguiculata (cowpea) BJMBR
Venâncio,T.M.; Oliveira,A.E.A.; Silva,L.B.; Machado,O.L.T.; Fernandes,K.V.S.; Xavier-Filho,J..
Since the discovery of bovine insulin in plants, much effort has been devoted to the characterization of these proteins and elucidation of their functions. We report here the isolation of a protein with similar molecular mass and same amino acid sequence to bovine insulin from developing fruits of cowpea (Vigna unguiculata) genotype Epace 10. Insulin was measured by ELISA using an anti-human insulin antibody and was detected both in empty pods and seed coats but not in the embryo. The highest concentrations (about 0.5 ng/µg of protein) of the protein were detected in seed coats at 16 and 18 days after pollination, and the values were 1.6 to 4.0 times higher than those found for isolated pods tested on any day. N-terminal amino acid sequencing of insulin...

Apparent cross-talk of two signaling pathways that regulate Zea mays coleoptile growth Phyton
Buentello Volante,B; Díaz de León-Sánchez,F; Rivera-Cabrera,F; Aguilar Caballero,R; Ponce-Valadez,M; Sánchez de Jiménez,E; Pérez-Flores,LJ.
Auxin and insulin promote Zea mays embryo growth, induce S6 ribosomal protein (S6rp) phosphorylation, and promote specific protein synthesis. The objective of this research was to test a possible cross-talk between insulin and auxin transduction pathways in Z. mays coleoptiles, typical auxin target tissues. Auxin and insulin produced differential quantitative and qualitative stimulation of cytoplasmic and ribosomal protein phosphorylation, and specific patterns of de novo synthesized cytoplasmic proteins. In addition, insulin induced S6rp phosphorylation was strongly inhibited by rapamycin, indicating target of rapamycin (TOR) kinase participation; auxin-induced S6rp phosphorylation was insensitive to this inhibitor. Phosphatidic acid (PA), a second...

Association of postalimentary lipemia with atherosclerotic manifestations BJMBR
Tentor,J.; Nakamura,R.T.; Gidlund,M.; Barros-Mazon,S.; Harada,L.M.; Zago,V.S.; Oba,J.F.; Faria,E.C. de.
We identified different lipemic and metabolic responses after the ingestion of a standardized meal by healthy adults and related them to atherosclerotic markers. Samples from 60 normolipidemic adults were collected before and after a liquid meal (40 g fat/m² body surface) at 0, 2, 4, 6, and 8 h for measurements of lipids, free fatty acids (FFA), insulin, cholesteryl ester transfer protein (CETP), autoantibodies to epitopes of oxidized LDL (oxLDL Ab), lipolytic activities, and apolipoprotein E polymorphism. Mean carotid intima-media thickness (cIMT) was determined by Doppler ultrasound. The volunteers were classified into early (N = 39) and late (N = 31) triacylglycerol (TAG) responders to the test meal. Late responders showed lower HDL cholesterol...

Biological response of spontaneously hypertensive rats to the streptozotocin administration BABT
Costa,Virginia Alice Vieira da; Vianna,Lucia Marques.
The sensitivity of adult spontaneously hypertensive rats (SHR) to the diabetogenic effect of streptozotocin (STZ) was studied. The animals were subdivided into three groups: control (citrate buffer), streptozotocin 40 mg/kg or 50 mg/kg, and general biologic parameters were analyzed, in addition to systolic blood pressure, blood glucose and insulin levels determinations. Both doses were able to induce hyperglycemia above 300 mg/dl; however, 50 mg/kg provoked a more pronounced physiological alterations in body weight, diuresis, water and food intake. There was no change on systolic blood pressure with either dose. Results suggested that SHRs did not need doses of streptozotocin above 40mg/kg in order to produce diabetes probably because this strain was much...

Biphasic modulation of insulin receptor substrate-1 during goitrogenesis BJMBR
Grozovsky,R.; Morales,M.M.; Carvalho,D.P..
Insulin receptor substrate-1 (IRS-1) is the main intracellular substrate for both insulin and insulin-like growth factor I (IGF-I) receptors and is critical for cell mitogenesis. Thyrotropin is able to induce thyroid cell proliferation through the cyclic AMP intracellular cascade; however, the presence of either insulin or IGF-I is required for the mitogenic effect of thyroid-stimulating hormone (TSH) to occur. The aim of the present study was to determine whether thyroid IRS-1 content is modulated by TSH in vivo. Strikingly, hypothyroid goitrous rats, which have chronically high serum TSH levels (control, C = 2.31 ± 0.28; methimazole (MMI) 21d = 51.02 ± 6.02 ng/mL, N = 12 rats), when treated with 0.03% MMI in drinking water for 21 days, showed...

Consequences of cerebroventricular insulin injection on renal sodium handling in rats: effect of inhibition of central nitric oxide synthase BJMBR
Oliveira,P.C.; Michelotto,J.B.; Zapparoli,A.; Boer,P.A.; Gontijo,J.A.R..
In the present study, we investigated the effects of acute intracerebroventricular (icv) insulin administration on central mechanisms regulating urinary sodium excretion in simultaneously centrally NG-nitro-L-arginine methylester (L-NAME)-injected unanesthetized rats. Male Wistar-Hannover rats were randomly assigned to one of five groups: a) icv 0.15 M NaCl-injected rats (control, N = 10), b) icv dose-response (1.26, 12.6 and 126 ng/3 µL) insulin-injected rats (N = 10), c) rats icv injected with 60 µg L-NAME in combination with NaCl (N = 10) or d) with insulin (N = 10), and e) subcutaneously insulin-injected rats (N = 5). Centrally administered insulin produced an increase in urinary output of sodium (NaCl: 855.6 ± 85.1 Δ%/min; 126 ng insulin: 2055 ± 310.6...

Diet plus insulin compared to diet alone in the treatment of gestational diabetes mellitus: a systematic review BJMBR
Giuffrida,F.M.A.; Castro,A.A.; Atallah,A.N.; Dib,S.A..
Fetuses of mothers with gestational diabetes mellitus are at increased risk to develop perinatal complications mainly due to macrosomia. However, in view of the marked heterogeneity of this disease, it seems difficult to set guidelines for diagnosis and treatment. This complicates the choice of assigning patients either to diet or to insulin therapy. Also of concern is how much benefit could be expected from insulin therapy in preventing fetal complications in these patients. In a systematic review of the literature assessing the efficacy of insulin in preventing macrosomia in fetuses of mothers with gestational diabetes, we found six randomized controlled trials comparing diet alone to diet plus insulin. The studies included a total of 1281 patients (644...

Calvo Arriaga, Ayne Oliva.

Effect of different levels of total dietary fiber on the performance of sows in gestation and lactation R. Bras. Zootec.
Oelke,Carlos Alexandre; Ribeiro,Andréa Machado Leal; Noro,Mirela; Bernardi,Mari Lourdes; Denardin,Cristiane Casagrande; Nunes,Paula Raymundo; Veit,Fernando Cézar; Winckler,João Carlos.
ABSTRACT The present study evaluated the effect of variation in the amount of total dietary fiber (TDF) in sow diets, provided from 74 to 114 days of gestation, on the digestibility of nutrients and energy during gestation, as well as productivity and metabolism during gestation and lactation. Thirty-three sows were used in a randomized experimental design. The amounts of TDF in three treatment diets were 156, 223, and 282 g kg−1. The daily intake of nutrients and energy was similar for all diets the entire period of gestation, differing only in the volume of ration consumed, which was 2.1, 2.2, and 2.4 kg day−1 (days 74 to 90) and 2.5, 2.6, and 2.8 kg day−1 (days 91 to 111) for the 156, 223, and 282 g kg−1 TDF diets, respectively. During gestation, the...

Effect of FSH and insulin on lipogenesis in cultures of Sertoli cells from immature rats BJMBR
Follicle-stimulating hormone (FSH) and insulin regulate glycide metabolism in Sertoli cells, thus stimulating lactate production. These stimulatory effects of FSH and insulin do not require protein synthesis, suggesting a modulation of enzyme activity and/or regulation of glucose transport. The present investigation was performed to characterize the hormonal control of lipid metabolism in Sertoli cells. The data indicate that FSH and insulin have a regulatory effect on lipid metabolism in Sertoli cells. After 8 h of preincubation with insulin (5 µg/ml), the activity of the enzyme ATP-citrate lyase in cultured Sertoli cells was increased from 0.19 to 0.34 nmol NAD+ formed µg protein-1 min-1. FSH (100 ng/ml) had no effect on this enzyme. Glycerol phosphate...

Effect of high-fat diets on body composition, lipid metabolism and insulin sensitivity, and the role of exercise on these parameters BJMBR
Coelho,D.F.; Pereira-Lancha,L.O.; Chaves,D.S.; Diwan,D.; Ferraz,R.; Campos-Ferraz,P.L.; Poortmans,J.R.; Lancha Junior,A.H..
Dietary fat composition can interfere in the development of obesity due to the specific roles of some fatty acids that have different metabolic activities, which can alter both fat oxidation and deposition rates, resulting in changes in body weight and/or composition. High-fat diets in general are associated with hyperphagia, but the type of dietary fat seems to be more important since saturated fats are linked to a positive fat balance and omental adipose tissue accumulation when compared to other types of fat, while polyunsaturated fats, omega-3 and omega-6, seem to increase energy expenditure and decrease energy intake by specific mechanisms involving hormone-sensitive lipase, activation of peroxisome proliferator-activated receptor α (PPARα) and...

Effect of intracerebroventricularly injected insulin on urinary sodium excretion by cerebroventricular streptozotocin-treated rats BJMBR
Macedo,R.F.; Furlan,F.C.; Marshall,P.S.; Michelotto,J.B.; Gontijo,J.A.R..
Recent evidence suggests that insulin may influence many brain functions. It is known that intracerebroventricular (icv) injection of nondiabetogenic doses of streptozotocin (STZ) can damage insulin receptor signal transduction. In the present study, we examined the functional damage to the brain insulin receptors on central mechanisms regulating glomerular filtration rate and urinary sodium excretion, over four periods of 30 min, in response to 3 µl insulin or 0.15 NaCl (vehicle) injected icv in STZ-treated freely moving Wistar-Hannover rats (250-300 g). The icv cannula site was visually confirmed by 2% Evans blue infusion. Centrally administered insulin (42.0 ng/µl) increased the urinary output of sodium (from 855.6 ± 85.1 to 2055 ± 310.6 delta%/min; N =...

Endocrine cells and nerve ganglia of the small intestine of the Opossum Didelphis aurita Wied-Neuwied, 1826 (Mammalia: Didelphidae) Anais da ABC (AABC)
Freitas-Ribeiro,Gláucia M.; Fonseca,Cláudio C.; Sartori,Sirlene S.R.; Loures-Ribeiro,Alan; Neves,Clóvis A..
The nervous and endocrine systems jointly control intestinal movements, secretions of their glands and also participate of the processes of nutrient digestion and absorption. Therefore, the central objective of this study was to verify the existence of a possible relationship between the number of nervous cells and ganglia of the submucosal and myenteric plexuses and the number of endocrine cells in the small intestine of adult D. aurita. The utilized staining techniques were Grimelius, modified Masson-Fontana, direct immunoperoxidase and H-E. Argyrophillic, argentaffin and insulin immunoreactive endocrine cells do not numerically vary between the initial, mid and final regions of the duodenum, jejunum and ileum (P>0.05), except for argyrophillic cells...
